Journal of Pharmaceutical Sciences: Evaluation of a dual-wavelength size exclusion HPLC method with improved sensitivity to detect protein aggregates and its use to better characterize degradation pathways of an IgG1 monoclonal antibody

Ultraviolet detection at two different wavelengths was utilized to monitor IgG1 monoclonal antibody levels of monomer, aggregate, and fragments and was shown to have improved sensitivity for the detection aggregates and fragments compared to light scattering (LS) detection. A TSKgel^® G3000SW_XL column was used in this technique.

In this study, the nature of morphological variation of amyloid ß proteins was examined by dark-field and transmission electron microscopy in a preparation of seeded amyloid ß protein fibrils that formed at relatively low protein concentrations and exhibited remarkably high thermodynamic stability. A TSKgel G3000SW_XL column was used in the preparation of the fibril seeds.

Saccharides are fundamental substances that express various bioactivities and may exist independently or form complexes with proteins or lipids. Sugar alcohols are a class of polyols and are commonly added to foods since they are of lower caloric content than the corresponding sugars. This note reports the successful analysis of different kinds of saccharides and sugar alcohols using a TSKgel NH₂-100 HILIC HPLC column.

SR109: TSKgel STAT Columns for High Performance Ion Exchange Chromatography

The high resolution and high throughput TSKgel STAT columns expand the range of applications for ion exchange chromatography. TSKgel NPR columns have been successfully used in the past to achieve high resolution in ion exchange chromatography of proteins and nucleic acids. The TSKgel STAT columns, which also use a non-porous packing material, provide the same or better resolution as the smaller particle size TSKgel NPR columns and as can be expected from their larger particle size, operate at lower column back pressure than TSKgel NPR columns. This 14 page Report discusses the basic characteristics of the TSKgel STAT columns and presents application examples.

Even at a low temperature of 5°C, column pressure is 10MPa or lower in each of these TSKgel STAT columns at a standard flow rate of 1.0mL/min. This clearly shows that these columns can be used even in low pressure LC systems.
