TSK-GEL SuperSW columns, introduced in 1997, represent the third generation of high performance Gel Filtration columns that have become synonymous with analyzing protein molecular weights in the emerging field of biotechnology.

Particle Characteristics
TSK-GEL SuperSW-type columns contain smaller particles than SWxl-type columns; 4 micron versus 5 micron. In addition, the column internal diameter has been reduced from 7.8mm ID to 4.6mm ID to provide higher sensitivity in sample-limited cases and to cut down on solvent use.

HPLC System Requirements
It is important to employ an HPLC system that is optimized with regards to extra-column bandbroadening to take full advantage of the high column efficiency that can be obtained on SuperSW columns. See Separation Report 95 for recommendations.

Calibration Curves
The figure to the right shows the calibration curves for globular proteins for the two TSK-GEL SuperSW-type columns in comparison with those for the corresponding SWxl columns. The following table summarizes the molecular weight ranges for SuperSW2000 and SuperSW3000 columns for globular proteins, polyethylene oxides and dextrans that can, at least partially, access the pores. For comparative purposes, similar data is provided as well for TSK-GEL SW and SWxl-type columns.

Columns:

Two 4µm, 4.6mmIDx30cm TSK-GEL SuperSW columns in series; two 5µm, 7.8mmIDx30cm TSK-GEL SWxl columns in series; two 10µm, 7.5mmIDx60cm TSK-GEL SW columns in series

Elution:

Globular proteins: 0.3M NaCl in 0.1M (0.05M for SWxl columns) phosphate buffer, pH 7
Dextrans and polyethylene glycols and oxides (PEOs): distilled water
